Transformations of folic acid with the participation of DHFR and MTHFR
As already mentioned, folic acid (a synthetic form present in most of the dietary supplements) undergoes a complex metabolism, which results in the production of the target bioactive form – tetrahydrofolate. Folic acid, with the help of dihydrofolate reductase (DHFR) and methylenetetrahydrofolate reductase (MTHFR) enzymes, is first reduced to dihydrofolate and then to tetrahydrofolate.

MTHFR gene mutation and absorption of folic acid
MTHFR refers not only to the enzyme but also to the gene responsible for its formation. In case of MTHFR gene mutations, the functionality of the enzyme encoded by the gene is disturbed, so is the metabolism of folic acid. This results in a significant reduction in the bioavailability of this compound, which may be the cause of folate deficiency, with many health consequences. Unlike folic acid, methyltetrahydrofolate, i.e. active folate, does not need to be reduced by MTHFR.

Folates and the homocysteine ​​level
Methyltetrahydrofolate is the source of one-carbon residues necessary in many metabolic processes, including in the transformation of amino acids. It is necessary for the remethylation of homocysteine ​​to methionine, which results in lowering the homocysteine ​​level in the body. Homocysteine ​​is a sulfuric amino acid, whose elevated level promotes the occurrence of cardiovascular diseases, disorders of the nervous system, including Alzheimer's disease and cancer (especially colon cancer).

Protection against anemia
One of the symptoms of folate deficiency is megaloblastic anemia, characterized by the presence of megalocytes – enlarged erythrocytes with shorter survival time and a slower production rate. This is due to the participation of folates and vitamin B12 in the proper formation of erythroblast cell nuclei. Symptoms of this type of anemia include easy fatigability, decreased energy levels, difficulty with concentration, headaches, and impaired fertility.

Folates – essential for normal pregnancy
The appropriate supply of folates is essential for the proper course of pregnancy. Folates are needed for proper divisions of intensely dividing cells, which include fetal tissue. Therefore, this is why the demand for folates increases during pregnancy. It is recommended to supplement folates already in the preconception period, as well as during pregnancy and after birth.

It has been shown that folate deficiency in the diet of a pregnant woman increases the risk of congenital malformations of the fetus, such as neural tube defects. The best solution seems to be the combined supplementation of active folate and folic acid (do not give up folic acid). The inclusion of methylated folate in the supplementation will bring benefits in the form of faster saturation of the body and better digestibility.

Folates for the health of the nervous system
Folates are essential for the proper functioning of the nervous system. There is more and more data on their neuroprotective properties. They participate in the synthesis of neurotransmitters, including adrenaline, noradrenaline, and dopamine. Numerous epidemiological studies indicate that folate deficiency contributes to the development of diseases and disorders of the nervous system such as depression, accelerated aging of neurons, dementia or various types of neuropathy. This is due to elevated levels of homocysteine, accompanying folate deficiency.
